Duolink® and Proseek® Protein Analysis Tools

The Duolink® products enable you to detect and individually count single endogenous proteins or single protein events, such as protein-protein interactions, protein dimerizations and post-translational modifications. Even transient or weak interactions can be assessed. These events can be accurately quantified in unmodified cells and tissues, starting with only a few cells.

Proseek® enables the accurate detection and quantification of proteins in a solution, such as serum and plasma, in a minimal sample volume, as low as 1 µl!

The Duolink® assay principle

Two primary antibodies raised in different species recognize the target antigen or antigens of interest. Species-specific secondary antibodies, called PLA® (Proximity Ligation Assay) probes, each with a unique short DNA strand attached to it, bind to the primary antibodies. When the PLA® probes are in close proximity, the DNA strands can interact through a subsequent addition of two other circle-forming DNA oligonucleotides.

After joining of the two added oligonucleotides by enzymatic ligation, they are amplified via rolling circle amplification using a polymerase. Detection can be performed in fluorescence or luminescence.

The Proseek® assay principle

The Proseek principle is based on the Proximity Extension Assay Technology (PEA): a pair of oligonucleotide-labeled antibodies (Proseek probes) are allowed to pair-wise bind to the target protein. When the two Proseek probes are in close proximity, a PCR target sequence is formed by a proximity-dependent DNA polymerization event. The resulting sequence is subsequently detected and quantified using standard real-time PCR.
