Oligonucleotide Synthesis and Quality Control

To coordinate and monitor the large synthetic projects, Eurogentec has designed and implemented the necessary database infrastructure to support high-throughput synthetic efforts. This laboratory information management system assists in oligonucleotide ordering, oligonucleotide annotation, tracking, and oligonucleotide quality control.

Quality Control


The following quality control metrics are applied to assess the quality and the quantity of the oligo.

1- Oligonucleotide synthesis are monitored by the trityl release following the first coupling and the last coupling. The step-wise repetitive yield is computed based on the total number of couplings.
2- Oligonucleotides with less than a 99 % coupling efficiency are rejected. After the final detritylation, the oligo is 5'-modified with a C6 amino-linker using standard phosphoramidite coupling chemistry. The oligonucleotides are subsequently deprotected, desalted, and the optical density of each amino-modified measured at 260 nm.
3- To monitor heterogeneity in oligonucleotide length, each oligonucleotide is subjected to control. The quality of the oligo’s will be controlled by the measurement of the molecular mass using MALDI-TOF Mass Spectrometry (MS). All the oligo showing a bad / poor MS profile will be re-synthesized at the expense of Eurogentec.

The sequence information of the designed probes is forwarded to our in-house oligo production team. 
In order to satisfy the demand of our customers, Eurogentec has established a high-throughput synthesis program capable for example of producing up to 500 5'-amino modified probes of 70 bases per day.

The amino-modified probes used for microarray fabrication are exhaustively analyzed to ensure probes are full-length and to guarantee the minimal yield of 4 nanomole per oligo.
