    Quality Control


    The S. lividans MicroArrays are certified to generate a high quality data.
    All printed slides are previously selected for coating uniformity and low intrinsic background.
    The MicroArrays are visually inspected for spotting quality (doughnuts), uniformity, integrity and for perfect alignment of spots and grids.
    From each batch of printed MicroArray, three are randomly chosen for further quality control. We use the Terminal Transferase reaction to assess the amount of DNA covalently attached to the slides. Finally, we perform a real hybridization using labeled cDNA obtained from heat-choc cultured cells.

    Development and production of a full long oligo DNA MicroArray representing the complete set of S. lividans / S. coelicolor genes was . The 7512 chromosome genes, the 351 SCP1 plasmids genes and the 33 SCP2 plasmids genes sequences were downloaded from the Sanger Institute website. In order to perform gene specific primer design of long oligos with the best possible thermo dynamical features for DNA array applications, the array designer 3.0 software commercialised by Premier biosoft was acquired. 7896 long oligos specific to the corresponding (putative) genes of S. coelicolor A3(2)(S. lividans) of a size of 68 bases were designed and produced at the Eurogentec’s oligo production facility. A complete set of DNA array specific controls such as positive, negative, dynamic range, spatial, normalization, hybridisation efficiency, reverse transcription efficiency and resistance genes controls were designed and produced
