TRI Insta-Pure: A three-step separation of RNA, DNA and proteins
TRI Insta-Pure is a complete ready-to-use reagent for the three-step isolation of RNA, DNA and proteins from tissues or cultured cells of human, animal, plant, yeast, bacterial or viral origin. The isolation method for RNA, DNA and proteins is built on the single-step liquid phase separation (RNA Insta-Pure, see above). This method has been shown to yield undegraded RNA from small and large samples. It also makes it possible to process a large number of samples simultaneously. TRI Insta-Pure includes phenol and guanidium thiocyanate in a monophasic solution. After addition of chloroform and centrifugation, the homogenate separates into three phases:
- Total RNA is isolated from the aqueous phase in 1 hour and is undergraded and free of proteins and DNA. It is ready for Northern and dot blotting, poly-A+ selection and reverse transcription. Yield: 1–15 µg RNA per mg tissue or 106 cells.
- Total DNA is isolated in 3 hours by selective DNA precipitation from the organic phase and is ready for restriction analysis, Southern and dot blotting, PCR and cloning. Yield: 2–7 µg DNA per mg tissue or 106 cells.
- Proteins are isolated in 3 hours and are ready for Western blotting.
