Several mutations in the beta amyloid precursor gene cause autosomal dominant Alzheimer's Disease in a number of kindreds. Among them, the Italian mutation (E22K) leads to beta-amyloid peptides which aggregate more rapidly and exhibit potent neurotoxicity compared to wild-type beta-amyloid 1-40. The formation of a salt bridge between Lys-22 and Asp-23 in the minor conformer might explain why E22K-beta-amyloid 1-40 is more pathogenic than its wild-type conterpart.
