Takyon® Ultra for RNA targets
Takyon® Ultra is an optimized One-Step RT-qPCR kit designed for RNA detection and precise gene expression analysis. It features an advanced reverse transcription Master Mix with enhanced multiplexing capabilities. This kit ensures sensitive and specific detection of RNA templates, including those from SARS-CoV-2, influenza, and RSV even in the presence of common qPCR inhibitors.

One-Step RT-qPCR Takyon® Ultra?
Takyon® Ultra is designed to support efficient and reliable one-step RT-qPCR workflows. It helps
researchers detect low-copy targets, handle inhibitor-rich samples, and simplify setup with a stable formulation.
Low copy detection
Our Master Mix formulation delivers clean non template controls (NTCs) and enhanced specificity, even in low copy viral target contexts, ensuring both reliable negative results and high detection sensitivity. In addition, Takyon® Ultra offers enhanced multiplexing capabilities, enabling the simultaneous detection of multiple targets with confidence.
Inhibitor tolerance
This Master Mix is designed to maintain robust performance in the presence of common qPCR inhibitors. Its resilient formulation ensures consistent amplification results across a wide range of sample types.
Increased stability
Thanks to the improved Euroscript II Plus reverse transcriptase, assays can be conveniently assembled at room temperature. It maintains consistent amplification at 20°C during 24 hours. This feature streamlines high-throughput workflows while ensuring sensitive and reproducible cDNA synthesis and amplification.

The One-Step Takyon® Ultra Probe 4X Master Mix (yellow) shows increased sensitivity compared to Competitor T (dark blue) and Competitor B (light blue) reagents, and increased specificity compared to competitor T, under their recommended fast cycling conditions.

Accurate multiplex detection of RNA
targets with Takyon® Ultra RT-qPCR
To demonstrate its suitability for RNA applications, Takyon® Ultra RT-qPCR was tested in a triplex
assay detecting three SARS-CoV-2 targets. The assay covered a dynamic range from 10⁵ down to
10¹ RNA copies, enabling direct comparison of amplification performance across different transcript abundances.

The data demonstrate that Takyon® Ultra RT-qPCR ensures robust multiplex amplification of RNA targets with high efficiency (>93%), strong linearity (R² ≥ 0.999), and excellent reproducibility, as confirmed by the consistent results obtained across technical replicates.
Improved formulation for a better resistance to inhibitors
The enhanced formulation of Takyon® Ultra increases its resistance to common qPCR inhibitors present in various sample types.

Takyon® Ultra One-Step Master Mix demonstrates enhanced resistance to heparin inhibition at concentration 2 IU/mL , 3 IU/mL and 4 IU/mL in RT-qPCR targeting Parainfluenza4.
Maintained performance in presence of sample treatment inhibitors

Takyon® Ultra One-Step Master Mix demonstrates enhanced resistance to Ethanol at concentration 1%, 3% and 5% in RT-qPCR targeting Parainfluenza4.

Takyon® Ultra One-Step Master Mix demonstrates enhanced resistance to humic acid inhibition at concentration 1 ng/µL, 4 ng/µL and 20 ng/µL in RT-qPCR targeting Corona virus E290.
Quantitative RT-PCR protocol
The One-Step Takyon® Ultra Probe 4X MasterMix produces consistent and sensitive results under Fast cycling conditions.
For increased convenience, a working blend of Master Mix and Euroscript II RT Plus can be prepared and stored at -20°C for up to one month.
This one-tube solution is also stable at 20°C for up to 24h allowing for time-consuming high-throughtput assay set-up.
Universal kit
With the included ROX reference dye supplied in separate tubes, the One-Step RT-qPCR Kit is compatible with all real-time PCR thermocyclers. It integrates seamlessly into any laboratory workflow.
